Tirzepatide: analytical profile for research buyers
A long, modified dual-agonist peptide — which means a more complex impurity profile and a higher analytical bar than short research peptides.
Part of the GLP-1 research peptides pillar.
What the molecule is
Tirzepatide is a 39-amino-acid modified peptide reported as a GIP/GLP-1 dual agonist. Like semaglutide, it carries a fatty-acid side chain that supports albumin binding; unlike semaglutide, the longer backbone multiplies the number of coupling steps and the potential for deletion and truncation impurities.
Reference molecular weight: ≈4813.5 g/mol for the parent compound. Confirm the salt form before interpreting MS data.
The analytical consequence of length
Each coupling cycle is rarely perfect. Over 39 residues, small per-step inefficiencies accumulate into a visible impurity envelope. Practical implications:
- Purification is harder and more expensive; a "98% pure" label at this length deserves a closer look at the chromatogram.
- Deletion sequences often elute close to the parent, so method resolution matters more than the reported percentage.
- A single peak with no visible shoulders on a generic gradient is a prompt to ask for the method, not a reason to relax.
HPLC method notes
Ask for column chemistry, dimensions, mobile phase, gradient slope and detection wavelength. At this hydrophobicity, small gradient changes shift retention and resolution; without the method, a purity number cannot be compared across suppliers. Also check whether the integration includes the impurity envelope or only the main peak.
MS and sequence confirmation
For a molecule this size, a single MS mass match is necessary but not sufficient: it confirms the expected mass, not the sequence. Where identity risk is high, MS/MS fragmentation or amino-acid analysis adds a layer. At minimum, the COA should state the expected mass calculation and show the observed spectrum with interpretable adducts.
Net content economics
At ≈4813 g/mol, a 10 mg fill is a small molar quantity, and water plus counter-ion fraction can be a meaningful share of the powder. Compare price per mg of net peptide, not per vial. A 10% content difference is a 10% price difference at constant quality — easy to miss when quotes are compared per box.
Handling
- Equilibrate sealed vials before opening; the cake is hygroscopic.
- Dissolve slowly; hydrophobic sequences wet slowly and are prone to foaming.
- Avoid vortexing; use low-binding tubes and pre-rinsed filters.
- Aliquot before freezing; repeated freeze–thaw is the usual cause of "it stopped working".
Verification checklist
| Item | What to look for |
|---|---|
| Identity | MS with expected mass calculation; MS/MS where risk is high |
| Purity | Chromatogram with method and full impurity integration |
| Content | Water content + net peptide content |
| Batch linkage | Lot number on vial, COA and invoice |
| Consistency | Two or three consecutive lots for reorder decisions |
Editor's take
Tirzepatide's length is its analytical cost. Buyers comparing purity percentages across suppliers should first confirm the methods were capable of resolving a 39-residue peptide.
Limitations
Molecular and analytical information only; no dosing, efficacy or human-use guidance. The batch COA governs the material supplied.
